Ed specifically mentions they change their relationships.
So in the trailer it looks like Scorpion and Sub Zero are brothers, and Mileena might be Kitana's bodyguard

Yeah. Scorpion has a Lin Kuei emblem on his costume, so there's definitely a closer relation between the 2 (I suspect former clan mates). I recall in the original timeline the Shira Ryu were originally a splinter faction of the Lin Kuei, and went with the yellow colour scheme to mock the Lin Kuei. That concept may be coming back.

and Mileena might be Kitana's bodyguard

This I hadn't considered, and that would be interesting. Mileena was created with Tarkatan ferocity to better guard Kitana.
